Thymosin beta-4 (Tbeta4) is a ubiquitous 43 amino acid, 5 kDa G-actin-sequestering polypeptide belonging to the beta-thymosin family; TB-500 is a synthetic peptide corresponding to the N-acetylated heptapeptide LKKTETQ, derived from the actin-binding domain of thymosin beta-4.


Research Background

Thymosin beta-4 and its actin-binding domain fragment have been examined across a range of in vitro cell culture systems, ex vivo tissue preparations, and animal models. In vitro studies have employed human umbilical vein endothelial cells (HUVECs), adult human cardiac myocytes, adult human cardiac fibroblasts, and chick aortic arch preparations to probe cellular responses at the molecular level. Animal research has included murine myocardial infarction models (permanent left anterior descending coronary artery ligation in C57BL/6J mice) in which systemic peptide administration was studied. Separately, pharmacokinetic and metabolite profiling of TB-500 — the synthetic LKKTETQ fragment — has been conducted in an equine model using liquid chromatography-mass spectrometry analysis of urine and plasma collected after a single administered dose, representing the first identification of TB-500 and its metabolites in post-administration biological samples from horses.

Structural studies using chemical cross-linking have shown that thymosin beta-4 contacts both the barbed and pointed ends of an actin monomer in an extended conformation, with lysine residues at positions 3 and 18 cross-linking to actin subdomains 3 and 1 respectively, and Lys-38 contacting subdomain 2 at the pointed end — a geometry proposed to sterically block actin polymerization and maintain a cellular reservoir of unpolymerized G-actin. Research on the seven-amino-acid actin-binding motif (LKKTETQ) has investigated its role in endothelial cell migration and vessel sprouting in vitro, with findings indicating that peptides lacking any portion of this motif were inactive in those assays and that the adhesion and sprouting activity observed was inhibited by the addition of exogenous soluble actin.
