The Recovery Stack groups BPC-157 and TB-500, two peptides that recur together throughout tissue-repair and regeneration research.

Why these compounds are studied together

Both compounds are studied within the same broad research theme — the biology of tissue healing. BPC-157, a stable gastric pentadecapeptide, has been investigated in in-vitro and animal models of tendon, muscle, ligament, and gastrointestinal repair, with research focused on angiogenesis and the nitric-oxide system. TB-500, a fragment of the actin-binding protein thymosin beta-4, has been studied for its role in actin regulation, cell migration, and wound-healing models. Their research interests converge on tissue regeneration, which is why the two are frequently examined within the same body of literature.

Individual research backgrounds

BPC-157

BPC-157 is a synthetic pentadecapeptide (a 15-amino-acid chain; sequence Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val, molecular weight approximately 1419.55 Da) corresponding to a partial sequence of a protein originally isolated from human gastric juice.

BPC-157 has been examined over approximately three decades of preclinical research, predominantly in rodent models (Sprague-Dawley rats, Wistar rats, mice, and genetically diabetic mice), as well as in beagle dogs and in vitro systems including cultured myenteric neurons and enteric glial cells (EGCs) derived from rat ileum and colon. In vitro studies have assessed its effects on neuronal survival rates and EGC proliferation in long-term culture, as well as on contractile responses in isolated human and rat intestinal muscle strip preparations. Limited early-phase clinical research has been conducted: a Phase I trial in 42 healthy volunteers was initiated in 2015 to evaluate safety and pharmacokinetics, and a Phase II study in ulcerative colitis reported no observed toxicity; no approved therapeutic applications exist, and no lethal dose has been established in toxicology studies.

TB-500

Thymosin beta-4 (Tbeta4) is a ubiquitous 43 amino acid, 5 kDa G-actin-sequestering polypeptide belonging to the beta-thymosin family; TB-500 is a synthetic peptide corresponding to the N-acetylated heptapeptide LKKTETQ, derived from the actin-binding domain of thymosin beta-4.

Thymosin beta-4 and its actin-binding domain fragment have been examined across a range of in vitro cell culture systems, ex vivo tissue preparations, and animal models. In vitro studies have employed human umbilical vein endothelial cells (HUVECs), adult human cardiac myocytes, adult human cardiac fibroblasts, and chick aortic arch preparations to probe cellular responses at the molecular level. Animal research has included murine myocardial infarction models (permanent left anterior descending coronary artery ligation in C57BL/6J mice) in which systemic peptide administration was studied. Separately, pharmacokinetic and metabolite profiling of TB-500 — the synthetic LKKTETQ fragment — has been conducted in an equine model using liquid chromatography-mass spectrometry analysis of urine and plasma collected after a single administered dose, representing the first identification of TB-500 and its metabolites in post-administration biological samples from horses.
